You won't need a car then. I'd walk down to the river, take a ride on the CityCat either up or down the river and back, then check out the Museum and GOMA precinct, and wander through Southbank to the Bridge that goes over to QUT. Walk through QUT to the Botanic Gardens and have a bit of an explore, and then walk back to the hotel.

Then you'll be in the heart of the city and walking distance to everything. You can easily walk to Southbank/Museum/Art Gallery/GoMA/QPAC and from there catch a city cat and cruise up and down the river if you want. Markets at Southbank, loads of restaurants. If you're into shows you could go and see Anything Goes (it's on til Sunday.. I loved it).

If you're staying in city I wouldn't worry about car hire. Public transport isn't great but there's enough to get you in and around CBD interesting things to see and do. CBD is open late Friday nights and walking bridge connects CBD with Southbank where there is lots to do.

Also there are markets at Riverside. From there you can catch the cross river ferry to Thornton St and walk along the bottom of the Kangaroo Point cliffs. It's an easy walk around to Southbank (although it will take you around 30-45 mins if you're taking an easy stroll). Lots to see and do.
Also, had a friend stay at that hotel recently. She said it was an awesome place to stay.

If your arriving from the airport if you can get a cab cheaper than $34 do that if not you can get a train as queen st isn't that far from a train station. If you do plan on using public transport use a go card as its cheaper than paper tickets since they don't do dailys or returns..

Take a city cat for a ride for as long as it goes and admire some riverside houses, and southbank, GOMA and queen St mall are all great and so much to see in walking distance from the hotel.
