I would either say something now or don't say anything. You can't eat the full packet of chips and then expect a refund because you were given chicken instead of the salt and vinegar that you ordered.

I'm not going to ask for a discount when I leave. I'll just mention that it was disappointing and they should consider disclosing it on the ad.

I don't think you are being very fair to the owners not to give them a chance to rectify the situation.
It may be that the cleaners haven't unlocked the third bedroom and the owners are not even aware that the house is not as you booked

this happened to us a couple of times. once overseas so I left a scathing review on trip advisor (we aired our issues to the owner who didn't seem overly fazed) and another time in noosa. I emailed the owners to give them a few pointers on why, when you request guests remove their own rubbish from the hotel room, you can't really call it 5 star luxury on your website. predictably my email was ignored. needless to say, I won't be returning.

definitely take it up with the owner. you've paid for what was described, why should you settle for less!?
Off topic but was this at RACV Noosa?

Yup! The key to unlock the door might even be in the house somewhere.

And if it's something this simple the OP will be kicking herself for not saying something sooner.

Only having 2 bedrooms isn't bothering me now the kids are coping with it. Arriving on the first night after travelling for 4 days it was annoying but really too late to ring the owner. The second day she was supposed to be round but I didn't see her and the kids fell asleep in the car on the way home from dinner so I didn't bother saying anything and after that I figured it would be rude to ask when is obviously managed for 2 nights.

I'm not going to ask for a discount, I'm not going to give a bad review, I'm just going to suggest that they should open all the bedrooms or disclose that they will leave some locked.

You're missing the point OP. Perhaps it was an innocent oversight in which case it serves no purpose (other than to point score/make the other party feel bad) if you bring it up at the end of the visit.

I agree. I would be mortified if a tenant waited until the end of a stay to say something that could easily be fixed.
